=== Types of vitamin A === It is "largely impossible" for provitamin carotenoids, such as beta-carotene, to cause toxicity, as their conversion to retinol is highly regulated. No vitamin A toxicity has ever been reported from ingestion of excessive amounts. Overconsumption of beta-carotene can only cause carotenosis, a harmless and reversible cosmetic condition in which the skin turns orange. In 1649, German scientist and inventor Otto von Guericke invented the spool vacuum air pump. Guericke's vacuum pump decreased any potential leaks between the piston and the cylinder by utilizing washers made from leather. In Britain, the first effective vacuum pump for scientific purposes was constructed in 1658 by English polymath Robert Hooke, on behalf of Anglo-Irish natural philosopher Robert Boyle. Boyle used the term "air pump" (among others) for his own vacuum pump as well as Guericke's, and they have often been referred to as such ever since. In 1705, English scientist Francis Hauksbee, developed a style of a double-barrelled air pump. === Protein === Due to alternative splicing, insulin-degrading enzyme has two isoforms. Isoform1 is ~118 kDa in size and composed of 1019 amino acids while the isoform 2 is ~54.2 kDa size and composed of 464 amino acids (missing 1-555 amino acids). Targeted mutations that favor the open conformation result in a 40-fold increase in catalytic activity. Based upon this observation, it has been proposed that a possible therapeutic approach to Alzheimer's might involve shifting the conformational preference of IDE to the open state, and thus increasing Aβ degradation, preventing aggregation, and, ideally, preventing the neuronal loss that leads to disease symptoms.
